Hiruko is the Black Samurai who came to the Eastern Kingdom of Mikado to sow chaos by distributing books to the Casualry class, causing them to transform into Demons and riot against the controlling Luxurors. She approaches Walter as the Black Samurai while he is investigating the incidents and tells him he will soon undergo change. Walter is unable to shake-off the meeting and sneaks into the monastery's storehouse to read one of the confiscated books, which prompts him to have a dream where the Black Samurai tells him to come to Tokyo.
When Walter arrives in Tokyo, the Black Samurai saves him from some Demons. Determined to know the figure's identity, Walter rips off the Black Samurai's helmet and discovers she is a dark-haired woman closely resembling Gina, his childhood friend and fellow Samurai. Hiruko introduces herself and leads Walter to her room in the district's underground to discuss matters. She explains that Tokyo is under control by the Ashura-kai's order and relates to the repression Walter had endured in Mikado. She attempts to persuade him to release his hatred and to join her in destroying the Ashura-kai. Their conversation is interrupted when Hiruko senses the arrival of Gina's presence in Tokyo, to her deep chagrin. Without telling Walter who, she explains that the presence is a threat to them but reassures that together they will prevail over it.
Hiruko tries to take Walter to Roppongi to eliminate the Ashura-kai together. When asked why she despises them, she deflects his question and states that the Ashura-kai's rule of order brings both humans and Demons suffering. She begins spiraling into madness and bleeds profusely as she vaguely recounts the abuse she had endured from the Ashura-kai. As Hiruko again asks Walter to accompany her, a group of panicked Demons burst into the room while fleeing in terror from a powerful entity, in actuality Gina. Walter flees from Hiruko in the midst of the confusion.
Hiruko later emerges from the rubble and kills a nearby couple, taking a woman's coat as she heads for the Sky Tower. There she begins massacring the Ashura-kai men gathered there before completely destroying the Sky Tower to prevent Walter from ever returning to Mikado. She then begins heading for Roppongi next.
At the same time, Walter reunites with Gina and uncovers her shared origins with Hiruko at the Reverse Hills in Roppongi: 12 years ago, the Ashura-kai had led a series of live research experiments aimed at creating a demon-human hybrid in an effort to pursue immortality. Their failed efforts had only produced defects that were quickly killed and disposed of. The project succeeded only after the archangel Abdiel lent his aid, which successfully produced an artificial angel named Gene. However, one of the defects who later became Hiruko, attacked and killed the researchers in retribution for their inhumane treatment. While Gene was transported by Abdiel to Mikado to protect her, where she became known as Gina, Hiruko had escaped and began living in Tokyo.
Hiruko locates Walter at Roppongi and saves him from being killed by the Ashura-kai. She brings him to shelter where she heals all of his wounds. However, Walter leaves her when he overhears people comment on a powerful demon slayer outside, believing this to be Gina. Hiruko follows Walter and comforts him when he finds Gina under the Ashura-kai's control and slaughtering Demons. Hiruko claims that she will never betray him and once again encourages him to join her in destroying their adversaries, suggesting that he becomes one with her. Walter agrees on the condition that their shared power be used to save Gina. Hiruko climbs into Walter's body through Amaymon's mouth, the Demon possessing him, and is devoured. The fusion transforms Walter into a humanoid Demon.
